Lena Luthor is the younger sister of Lex Luthor and is a scientific genius. She applied for Super Hero High School but was rejected after failing the psych test and took it really hard. Lex Luthor blamed Super Hero High. Fueled by his desire to see his sister become the world's greatest super, Luthor rationalized if he removed the top students from the school's roster, Super Hero High would become so desperate for new students that they would accept his sister. She was unaware of his plans. He utilized a suit she built equipped with stealth technology, endowed its user with high speed, reactive movements, and included infrared night goggles. For his first mission, Luthor vowed to take down the top supers in Metropolis. He targeted Supergirl, Wonder Woman, Bumblebee, Katana, Poison Ivy and Batgirl the day before final exams. They banded together in his Kryptonite powered prison and overcame him. He vowed his sister would get revenge.
The next semester, Lena Luthor was hired by Super Hero High to be its new I.T. girl after the post was vacated by Barbara Gordon. During a full moon, Wonder Woman and Supergirl worked on their shop projects well into the night. Doc Magnus informed them he was going to close up soon and reminded them they could come back in the morning. Lena Luthor arrived with the copper wiring Magnus requested and noticed Supergirl and her rocketship. She admired the ship. Supergirl noted it was her parents that built it and she was only adding mods to it. Luthor noticed gunk on the hull and wiped it off. Supergirl mentioned dust from Krypton crystallized and stuck to everything. Just as Supergirl welcomed her to Super Hero High, she picked up Batgirl's emergency signal and took off to rescue her from Killer Moth. After Supergirl left in her rocketship, Luthor remarked it was nice to meet her. She took the dust back to her secret lab and studied it. Luthor decided to resolve the weak potency of the sample and used an eye dropper to add a solution to it.
Luthor marveled at the creation of her first Kryptomite and was about to take a selfie to mark the milestone but she got a proximity alert. Supergirl, Wonder Woman, and Batgirl arrived. Luthor rushed upstairs to greet them. Supergirl explained she accidentally blew up her I.T. office. Luthor shrugged it off and hurried them outside. She returned to the lab and saw a huge mess. To make matters worse, the Kryptomite was gone. She went back to work. Donning a hazmat suit, she synthesized a Red Kryptomite and released it into the city.
Luthor partnered with Eclipso to harness the Earth's electromagnetic energies for evil. After stealing several generators from S.T.A.R. Labs and appropriating them, Luthor planted three devices on three of Earth's greatest localized hot spots for the energy: Egypt, the Aztec Jungle, and Stonehenge. The generators increased the flow of the electromagnetic energy at an exponential rate, endangering the planet. Luthor was told the energies were going to be used to generate a kind of hyperspace wormhole to Gemworld so Eclipso could return and exact revenge for being banished. In return, Luthor would be left the devastated planet to rule. In reality, Eclipso was using the generators to realign Earth's electromagnetic energy patterns with Gemworld to be in sync then transfer the organic matrix and terraform Earth into a new Gemworld to rule. She secretly had her Purple Kryptomites build a fifth generator at her palace on the Moon. Luthor planted special breeds of Kryptomite at each location and a fourth and much larger generator on Themyscira as a failsafe in case the other three were destroyed.
As she predicted, the three generators were destroyed by Wonder Woman, Batgirl, and Supergirl despite planting Kryptomites with specific powers to combat them: Yellow, Blue, and Orange respectively. She flew to Themyscira in her hovercraft only to discover the fourth generator was already destroyed. Eclipso arrived in her spaceship and announced her true plans then returned to the Moon. Lied and betrayed, Luthor allied with the girls to take down Eclipso's generator. Supergirl flew Batgirl and Luthor to one of the underground Batbunkers to create a computer virus to take down Eclipso's machine. Luthor was unable to open much less find a backdoor into the generator's software. Supergirl flew the virus to the machine herself then took them back to Themyscira where victory was celebrated. Supergirl wondered what would come of Eclipso. Luthor correctly mused the Purple Kryptomites would turn on her and hypnotize her.
Luthor figured out how to combine every type of Kryptomite into one and named it Rainbow Kryptomite and planned to take over the world with an army of them. From her hovercraft, Luthor dropped a rainbow pod into the fountain outside Super Hero High. As the Kryptomites swarmed Metropolis, Luthor savored victory at Capes & Cowls Cafe. Batgirl and Bumblebee assembled a giant jet from pieces of the Batjet and Cyborg then chased down Luthor. They blew up the fountain and destroyed the Kryptomites. Luthor discovered the Flash in her lab and strapped a Blue Kryptomite to his back. He ran around the city in confusion until Bumblebee, Supergirl, Batgirl, and Wonder Woman immobilized him with a grapnel line and separated the Kryptomite. Luthor soon finished a machine that could take away any supers' powers. She plotted to use it on Super Hero High and teamed up with Killer Moth.
Batgirl disguised herself as Supergirl and attempted to infiltrate Luthor's lab but she was quickly caught. Luthor demanded to know all the supers' weaknesses so she could finish her machine. Batgirl informed Luthor and Moth that Wonder Woman had a ticklish left elbow and Bumblebee was allergic to otters. While they conferred in private, Batgirl used her concealed rocket pack to burn through her bonds and turn off the machine. The real Supergirl arrived and destroyed the machine. Batgirl tied up Luthor, Moth, and a Green Kryptomite.
