S.T.A.R. Labs, short for The Scientific and Technological Advanced Research Laboratories, is a private research and development firm originally established near Metropolis. Founded by Garrison Slate, STAR Labs became the forefront of scientific research and paved the way for breakthroughs in various fields such as medicine, biology, physics, technology, engineering, and recently, metahumans. Today, S.T.A.R. Labs maintains facilities across the United States and the world, including Metropolis, Gotham City, and Paris.

When Batman first encountered Scarecrow, he eventually ran a search for known companies in Gotham City who specialize in chemicals that Scarecrow would incorporate in his Fear Toxin. The Gotham branch of S.T.A.R. Labs came up as one of the search queries along with Scarcrow's own company, Crane Chemicals, Axis Engineering, Bio Laboratories, and Macurity Manufacturing. Years later, Superman and Lois Lane met with Professor Hamilton at the central branch of S.T.A.R. in Metropolis with a sample of Kryptonite. Hamilton studied its properties and deduced its weakness to lead. A short time later, the Labs' janitor Rudy Jones attempted to aid in a theft to pay off his gambling debts. In the escape Jones was doused in chemicals designed to catalyze energy absorption in human cells. He survived the exposure and was mutated into Parasite. Parasite used the maintenance access in S.T.A.R. as his hideout while draining Superman of his power. Jimmy Olsen stumbled upon it and saved Superman.

In response to their connection to Parasite, a temporary holding cell was created in the Labs to house him. Afterwards, S.T.A.R. began to study metahumans. Their research was expedited when Superman entered into a reciprocal relationship. In exchange for studying the lifeboat Superman came to when he was a child, S.T.A.R. reverse engineered Kryptonian technology and other gagdets which they fully shared with Superman. Some of the objects made were an Anti-Kryptonite Suit, Space Suit, Scuba Suit, a second Phantom Projector, and a modified Lifeboat. They also confiscated dangerous objects such as weapons grade Kryptonite and a Mother Box and kept them in specially sealed vaults. They also solved problems that not even Superman could, such as stopping the growth spurts of the test monkey, Titano, and saved the life of an injured Supergirl. After the surgery on Supergirl was a success, relations between S.T.A.R. and Superman were strained due to the recent Apokoliptian invasion.

Years later, advance agents of the Imperium occupied an abandoned S.T.A.R. Labs facility. Two and a half years later, Batman discovered the nest and freed several humans kept in stasis. However, he was knocked unconscious by a sentry. Luckily, he activated a hypersonic watch and was saved by Superman. Some time later, Kaznian elite guard were assigned to steal a piece of classified weapons technology, a targeting device, from the Paris branch of S.T.A.R., but were met by Batman. Batman managed to apprehend one of the squad and interrogated him in the facility.

After the formation of the Justice League Unlimited, relations appeared to be formed with S.T.A.R. but beneath the surface, it was allied with Cadmus. Professor Hamilton fed a convincing story to Supergirl, the Question, and Green Arrow when they arrived at the Metropolis branch to investigate Supergirl's nightmares. Upon leaving the facility, the trio was attacked by Z-8 Trainer synthoids and Cadmus soldiers. S.T.A.R. and Hamilton willingly played along in a plan hatched by Lex Luthor to publicly humilate Superman and the League. Hamilton alerted the League of a theft of weapons-grade Kryptonite from the Labs. Superman and Batman investigated and soon after when Superman discovered a Kryptonite power generator under Lexor City, he would suspect foul play and try to dismantle it before the countdown expired. However, it was all a simple power source.
