Dr. Harleen Quinzel was Hugo Strange's assistant. On the night of the Evil Extractor demonstration at the Gotham State Penitentiary's Top Secret Prison Lab, Quinzel blushed after the Joker winked at her. After the super criminals were all strapped into their seats, Quinzel gave a wink back to Joker. Strange directed her to instigate the extractor protocol. Quinzel decompressed the cerebral defibrillator and pushed a lever up on the console. On Strange's call, she adjusted the hemostatic stabilizers then pushed up the field discharge switch. Strange realized the super criminals were trying to overwhelm the machine with their evil and yelled at Quinzel to shut down the Evil Extractor. Quinzel pushed the discharge switch and the Extractor powered down but the alarm kept blaring. District Attorney Harvey Dent was doused with Evil Essence that leaked out of the Extractor's Collection Vent and transformed into a new super criminal named Two-Face. Quinzel remarked it looked like that smarted. Strange was fired soon after.
Over half a year later, Quinzel took on her own super criminal persona, Harley Quinn, based on the harlequinn. She sneaked into the Gotham State Penitentiary and blew up the Joker's glass enclosure with a claymore bomb. When Joker looked up, Quinn held her oversized mallet and proclaimed, "Knock, knock, puddin'. Say hello to your new and improved Harley Quinn."
