Drury Walker, while in prison with an identification number 234026, read an article about Batman. Walker decided to reverse his unsuccessful criminal career by becoming the antithesis of Batman, a Killer Moth. After being released from prison, Walker used his stolen funds to construct a Mothcave and Mothmobile based on alleged photos of the Batcave and Batmobile.

With his remaining funds, he created an alias and back-story; Cameron van Cleer, millionaire philanthropist. Moth was able to become friends with the Gotham elite, including Bruce Wayne and gifted criminals with a Moth-signal. On his first outing, Killer Moth rescued several criminals from the police and defeated Batman. A rematch took place on the Gotham Bridge and Killer Moth escaped by diving 1000 feet into the Gotham River.

Or at least, that's the story that's told on the streets. Killer Moth is considered a third tier super villain and is more of an annoyance, even by fellow villains. After hearing about Team Penguin, Killer Moth tried to gain membership because of his regard of the Penguin. Moth was delegated to team driver. While delivering an unstable chemical, Moth mutated into a giant moth beast. His personality still remained intact. The Penguin tried to use him, alone, to break into Robert Howard's home and steal a ruby-encrusted penguin only to be defeated by the Bat Family.
